The same in vitro study reported that the GHRP-1–induced GH release was not affected by a GRF antagonist ([Ac-Tyr1, D-Arg2]-GRF 1-29). Additional review-style sources state that GHRPs act via specific receptors at the pituitary and/or hypothalamic level, are non-structurally homologous to GHRH, and that the mechanism remains unclear, with hypotheses involving counteracting somatostatinergic activity and/or GHRH-mediated mechanisms, including the possibility of an unknown hypothalamic factor.
